Show 65 dosing solutions, respectively. Bond Elut Certify™ solid phase extraction columns were obtained from Varian (Harbor City, CA). Methanol and ethyl acetate (HPLC grade) were obtained from Burdick and Jackson (Muskegon, MI). Ultra-high purity helium (99.999%) was obtained from Mountain Airgas (Salt Lake City, UT). All drug solutions used for quantitative analysis were prepared in HPLC grade methanol and HPLC grade solvents were used in all extractions. RIA analysis of mouse plasma samples was performed using a Coat- A-Count PCP RIA kit purchased from Diagnostic Products Corporation (Los Angeles, CA). AHP, 57% HI, 50% hypophosphorus acid, 3% hydro gen peroxide and octane sulfonate were obtained from Aldrich Chemical (Milwaukee, WI). Sepia melanin was purchased from Sigma Chemical (St. Louis, MO) Soluene-350 was purchased from Packard (Meriden, CT). AHP and sepia standard solutions were prepared in Milli-Q grade water. All other reagent grade chemicals were obtained from Mallinckrodt Chemical Works (St. Louis, MO). Animals Male SD, DA, LBNFl' LE, CP rats (-150 g), SW and C57BL6 mice (-35 g) were obtained from Harlan-Sprague Dawley (Indianapolis, IN). Male C57BL6 AY/a mice (-35 g) were obtained from Jackson Laboratories (Bar Harbor, ME). Animals were maintained in an environmentally controlled room with a 12-hour light-dark cycle and free access to food and water. Rats (n = 5, each strain) were individually housed in hanging wire cages to prevent contamination from urine or saliva from other rats or the bedding during the hair collection experiment. Mice were housed, 3 per box, in boxes lined with Dri-Dek® flooring (Naples, FL). To control for any potential contamination from the mice interacting with each other, a control mouse was included in each box (dosed: n = 7 for SW and C57BL6, n = 5 C57BL6 AY/a; control: n = 4, each strain). The control mice were not dosed, but hair was collected and analyzed for PCP as described below. |
