Show 44 Methods Chemicals and reagents PCP and phenobarbital (1 mg/ml in methanol) and PCP-ds and phenobarbital-d, (100 ug/rnl in methanol) were obtained from Radian Corp. (Austin, TX) for preparation of standard curves. PCP, phenobarbital (1 mg/ml in methanol), PCP-HCI and phenobarbital powder were obtained from Sigma Chemical (St. Louis, MO) for preparing quality control samples for accuracy evaluation and for preparation of dosing solutions, respectively. Bond Elut Certify™ solid phase extraction columns were obtained from Varian (Harbor City, CA). Methanol, methylene chloride, isopropanol and ethyl acetate (HPLC grade) were obtained from Burdick and Jackson (Muskegon, MI). Ultra-high purity helium (99.999%) was obtained from Mountain Airgas (Salt Lake City, UT). Tetramethylammonium hydroxide (TMAH) was obtained from Alltech Applied Science (State College, PA). All other reagent grade chemicals were obtained from Mallinckrodt Chemical Works (St. Louis, MO). All analytical drug solutions were prepared in HPLC grade methanol and HPLC grade solvents were used in all solid phase extractions. Tritiated PCP (12.68 CiJmmol) was obtained from RTI (Research Triangle Park, NC). Sepia melanin was purchased from Sigma Chemical (St. Louis, MO). ScintoSafe scintillation cocktail was purchased from Fischer Scientific (Pittsburgh, PA). A Coat-a Count barbiturate radioimmunoassay (RIA) kit was purchased from Diagnostic Products Corporation (Los Angeles, CA). Animals Male LE rats (-150 g) were obtained from Harlan-Sprague Dawley (Indianapolis, IN) and maintained in an environmentally controlled room with a 12-hour light-dark cycle and free access to food and water. Animals were housed individually in wire hanging cages to prevent contamination from the urine or saliva from other rats, or from the bedding.
