Description Non-specific alkaline phosphatase, ATP-ase, and glucose-6-phosphatase activities were assayed in intact tissue cultured cell from monkey kidneys with suitable substrate-buffer systems. The results were expressed in terms of released orthophosphate, over a give time, from cells in contrast with the sodium salts of glycerophosphate, ATP, or glucose-6-phosphate. The reproducibility of results was verified in each experiment in multiple analysis on a number of cell monolayers treated in an identical fashion. The effect of substrate and substrate concentration, pH, Ca++ and Mg++ concentration, glucose and L-glutamine, maleic acid, and 2,4-dinitrophenol on certain of these enzyme activities was determined. Possible variations in enzymatic activity between normal and poliovirus infected cells were noted, and the capacity of the cells to synthesize virus was measured under each of the assay conditions used. Factors which interfered with normal viral synthesis were reproduced with cells in a nutritionally complete medium, in certain instance, to see if the original interference still persisted, or to find whether the amount of virus produce in a given time was altered from normal tissue culture condition. The results from these experiments are summarized in the Discussion of this thesis.
