Description Melanopsin expressing Intrinsically-photosensitive-Retinal-Ganglion-Cells (ipRGCs) survive late in dominant optic atrophy (DOA), long after other RGC sub-types (and vision) have been lost. We hypothesise that specific neuroprotective genes, or pathways, are upregulated in ipRGCs transcriptomes compared to other RGC subtypes, affording them this resistance. These genes could, therefore, be harnessed to develop novel therapeutic strategies. Purpose (i)To evaluate a pathway for the preparation and enrichment of single cell suspensions of RGCs from mouse retina. (ii)To compare gene expression between RGC subpopulations (including ipRGCs) in a model of DOA using single cell RNA sequencing (scRNAseq).
