Description In this dissertation I present the contributions that I have made to two separate projects, each concerning different aspects of DNA replication in the yeast Saccharomyces cerevisiae. One project is a study of the Ctf18 protein, a member of the Replication Factor C (RFC) family of DNA polymerase processivity clamp loaders. I discuss the significance of the homology between Ctf18 and RFC, and a putative role for Ctf18 in an alternative" RFC complex. The second project concerns the role of Sptl6 and Pob3, which act together as both a transcription factor and as DNA replication factor, apparently by modulating the properties of nucleosomes.
