Cloning of DNA from Datura Innoxia into Charon 4A phage

DNA was isolated from Charon 4A phage and from suspension cultures of wild type Datura innoxia (WDI) plant cells. The DNAs were restriction cut and appropriate lengths for cloning were isolated and ligated. The hybrid DNA was packaged in vitro and the resultant clones were grown on bacterial plates. WDI RNA was also isolated and hybridized to Southern blots of WDI DNA which had been restriction cut and electrophoresed. From this a mpa of the WDI rRNA was determined.
